Katie - May I suggest that you obtain a copy of the publication MUSEUM MISSION STATEMENTS: BUILDING A DISTINCT IDENTITY. The publication is full of case studies about museums that revised their missions, often as part of strategic planning. It is available for $20 from the American Association of Museums as one of the Professional Practice Series. You may be able to borrow a copy from a local institution.
